چکیده

Male adult rabbits were divided into five experimental groups and were fed a 2% cholesterol diet using the following schedule: (A) two groups, normal control diet; (B) one group, 60 days continuous diet (60 day); (C) one group, 30 days cholesterol 30 days control diet (3030); (D) one group, 60 days cholesterol, 30 days control diet (60-30). Aortic collagen synthesis was estimated by measuring tissue prolyl hydroxylase activity and tissue cholesterol was measured following thin layer chromatography. In the 30-30 and 60-30 animals, the removal of cholesterol from the diet caused a rapid fall in serum cholesterol values which slowed the rate of aortic cholesterol accumulation. Collagen synthetic activity was significantly elevated in the 60-30 rabbits but was at control levels of both the 60 day and 3030 rabbits. The results demonstrate that the onset of increased aortic collagen synthesis is accompanied by an increase in the rate of cholesterol accumulation.

Lesion regression and protein synthesis in rabbits after removal of dietary cholesterol.

This study was conducted to determine whether atherosclerotic lesions regressed in rabbits that were placed on a normal, low cholesterol diet. Rabbits (n = 16) were fed a 2% cholesterol diet for 90 days; nine rabbits were killed, and the remaining seven rabbits were fed a low cholesterol diet for 7 months.. Appropriate noncholesterol-fed controls were used with each group. After 90 days of chol...

Changes in aortic lysyl oxidase activity in diet-induced atherosclerosis in the rabbit.

This study assessed the responses of lysyl oxidase, the enzyme that initiates covalent crosslinking in elastic and collagen, by studying the aortic tissue of rabbits after arteriosclerosis had been induced by diet. Rabbits in the experimental group were fed an atherogenic diet of rabbit chow supplemented with 8% peanut oil and 2% cholesterol for varying periods of time, while the control group ...

Synthesis of prostaglandins and thromboxane B2 by cholesterol-fed rabbits.

Alterations in the synthesis of thromboxane A2 (TxA2) and prostacyclin have been implicated in the development of atherosclerosis. We measured the amounts of the degradation products of these substances, TxB2 and 6-oxo-prostaglandin F1 alpha (6-oxo-PGF1 alpha), respectively, as well as PGE2, that were synthesized by slices and the luminal surfaces of aortas from rabbits fed either a control die...

Icaritin Inhibits Collagen Degradation-Related Factors and Facilitates Collagen Accumulation in Atherosclerotic Lesions: A Potential Action for Plaque Stabilization

Most acute coronary syndromes result from rupture of vulnerable atherosclerotic plaques. The collagen content of plaques may critically affect plaque stability. This study tested whether Icaritin (ICT), an intestinal metabolite of Epimedium-derived flavonoids, could alter the collagen synthesis/degradation balance in atherosclerotic lesions. Rabbits were fed with an atherogenic diet for four mo...

The effect of agents interfering with soft tissue calcification and cell proliferation on calcific fibrous-fatty plaques in rabbits.

We tested agents that might suppress the formation of fibrous plaques which are the significant lesions in human atherosclerosis. Six groups of rabbits were studied for 8 weeks: one group was fed a control diet and five groups received an atherogenic diet known to produce accumulations of connective tissue in arterial lesions. One group received only the atherogenic diet and the remaining four ...
